TITLE AND BRIEF PROJECT DESCRIPTION:Uncovering novel targets and pathways to enhance the efficacy of combination immunotherapy for lung cancerKMN309-CUP: E23C24000220006 Immunotherapies with immune checkpoint inhibitors (ICIs) have revolutionized the treatment of solid tumors, including lung cancer. Combining ICIs with conventional chemotherapy has proven successful, but only in a subset of patients. Although ICIs reinvigorate dysfunctional T cells against the cancer, acquired resistance occurs due to additional suppressive mechanisms, such as alternative checkpoints, inhibitory cell populations, and insufficient inflammation, among others. However, knowledge in this area is limited. Using a multitask approach that combines state-of-the-art technologies with the advantages of genetically engineered mice, models of primary carcinogenesis, and investigations in human samples, we will unravel the complex relationships between the immune system, the stroma, and the tumor. Our aim is to discover major cellular and molecular mechanisms of resistance, which will be further explored in preclinical models of lung cancer to enhance tumor regression.
